Can I play BDMV Folder using Cyberlink PowerDVD directly? without converted to ISO

If Can't, How to play BDMV Folder at Windows PC ? using what software ?

Simple, you can use VLC to play BDMV folder. Just select Media menu, go to "Open Disc...", under "Disc Selection" select "Bluray" and under "Disc Select", browse for the Blu ray folder containing BDMV and Certificate. For more help, you can use google to find the necessary information on how to further playback.

There's also Leawo Blu-ray Player. It's not great, but it gets the job done. You can simply drag and drop folder, or the MovieObject file in the bdmv folder.
